However, carbohydrates are difficult to analyze by using common chromatography methods since they share similar polarities and structures. High-performance anion-exchange chromatography with Pulsed Amperometric Detection(HPAE-PAD) is the main method we used to detect carbohydrates. ESI-MS and TOF are also used for carbohydrate analysis to meet extra requirements.

Methods and Instruments

HPAE-PAD detects carbohydrates by their electrical current generated by their oxidation at the surface while PAD will greatly enhance the sensitivity. Meanwhile, if the sample has a relatively high polarity and is thermally unstable, ESI-MS which dissolves sample into aerosol will be a better way to analyze molecules with similar polarities and structures.
